The aim of the present study was to investigate the clinical pharmacokinetics of ciprofloxacin, a single dose of ciprofloxacin, in healthy volunteers.
A total of 200 healthy volunteers participated in this pharmacokinetic study. Blood samples were collected using a standardized procedure. The concentrations of ciprofloxacin in serum were determined using a validated liquid chromatography-tandem mass spectrometry method, as previously reported for the pharmacokinetic studies in healthy volunteers [].
After the administration of ciprofloxacin (25 and 100 mg) into the rat, a single dose of ciprofloxacin (0.1 and 1 mg/kg) was administered orally (0.125-mg) for 14 days. The study included 48 healthy male volunteers. After the single dose of ciprofloxacin, blood samples were collected at three intervals (4, 12, 24 and 48 h), and the concentrations of ciprofloxacin were determined using a validated liquid chromatography-tandem mass spectrometry method. The mean concentration of ciprofloxacin in plasma was estimated to be 0.6 ng/mL (median: 0.5 ng/mL), and the mean concentration of the plasma protein binding was found to be 3.1 ng/mL (median: 5.0 ng/mL).
After ciprofloxacin administration, the mean concentration of ciprofloxacin in plasma was found to be 1.6 ng/mL (median: 1.4 ng/mL). The concentration of ciprofloxacin in urine was found to be 0.6 mg/mL (median: 0.7 mg/mL). The mean concentration of ciprofloxacin in plasma was 3.5 mg/mL (median: 3.0 mg/mL). The mean concentration in faeces was estimated to be 1.4 ng/mL (median: 1.3 ng/mL).
After oral administration of ciprofloxacin, the plasma levels of ciprofloxacin were found to be within the upper reference range, with a mean value of 4.5 ng/mL and a median value of 2.3 ng/mL. The plasma levels of ciprofloxacin were found to be within the lower reference range, with a mean value of 0.7 ng/mL and a median value of 2.6 ng/mL. The mean values of the pharmacokinetic parameters for ciprofloxacin in plasma were found to be within the upper reference range, with a mean value of 1.4 ng/mL and a median value of 2.6 ng/mL. The pharmacokinetic parameters for ciprofloxacin in urine were found to be within the upper reference range, with a mean value of 1.2 ng/mL and a median value of 3.0 ng/mL.
The mean plasma levels of ciprofloxacin were found to be 3.7 and 2.4 ng/mL, respectively, in healthy volunteers.
In a single dose of ciprofloxacin, the mean concentration of ciprofloxacin in the urine was found to be 0.8 ng/mL (median: 1.3 ng/mL), and the mean concentration of the plasma protein binding was found to be 1.3 ng/mL (median: 0.7 ng/mL). The mean values of the pharmacokinetic parameters for ciprofloxacin in urine were found to be within the upper reference range, with a mean value of 1.2 ng/mL and a median value of 2.6 ng/mL.
After oral administration of ciprofloxacin, the mean concentration of ciprofloxacin in plasma was found to be 1.3 ng/mL (median: 1.3 ng/mL). The mean values of the pharmacokinetic parameters for ciprofloxacin in plasma were found to be within the upper reference range, with a mean value of 1.6 ng/mL and a median value of 2.6 ng/mL.
In a single dose of ciprofloxacin, the mean concentration of ciprofloxacin in the urine was found to be 0.9 ng/mL (median: 1.4 ng/mL), and the mean concentration of the plasma protein binding was found to be 1.1 ng/mL (median: 0.6 ng/mL).
